چکیده

The oxidation of 3-hydroxyanthranilic acid, an intermediate in the conversion of tryptophan to niacin (l-3), is catalyzed by a liver oxidase (4). The first recognized product, termed Compound I (5), yields quinolinic acid spontaneously or picolinic acid under the influence of a liver carboxylase (6). The intermediate is detectable by its absorption maximum at 360 rnp (4). Wiss and Bettendorf (7) have identified the compound as I-amino-4-formyl-l , 3-butadiene-l , 2-dicarboxylic acid on the basis of infrared and ultraviolet spectra of several derivatives. Other studies in this laboratory (8) are directed towards obtaining further information concerning the structure of Compound I. In the kinetic studies to be reported the rate of appearance of Compound I has been used as a measure of oxidase activity. Activation of dialyzed, aged enzyme extracts by ferrous iron and reduced glutathione has been observed and inhibition by sulfhydry1 reagents has been studied.

Studies on purified 3-hydroxyanthranilic acid oxidase.

3-Hydroxyanthranilic acid oxidase is known to cat, alyze the oxidation of 3-hydroxyanthranilic acid to a very unstable aliphatic compound, which may either be spontaneously transformed into quinolinic acid or undergo enzymic conversion to picolinic or nicotinic acid, or both (l-3). According to Mason (4), this enzyme has been classified as an “oxygen transferase,” since the oxygen consumed, 1 mo...

1. 3-Hydroxyanthranilic acid oxidase has been purified from rat and ox liver. The most active preparations of the enzyme oxidize 2500 ,um-moles of 3-hydroxyanthranilic acid/min./mg. of protein. 2. The dependence of the reaction rate on the concentration of both substrates (3-hydroxyanthranilic acid and oxygen) was measured. Km for 3hydroxyanthranilic acid is low (7 KM under the conditions of th...

Effect of 3-hydroxyanthranilic acid on the mitochondrial respiratory system.

Studies on the metabolites of tryptophan in urine, in various neoplastic diseases, have shown an unusual excretion pattern, some of these compounds being eliminated in greater amounts than in normal human beings (Boyland & Williams, 1956a, b; Musajo, Benassi & Parpajola, 1955; Quagliariello, Tancredi, Fedele & Saccone, 1961; Tompsett, 1959; Ivanova & Ramonova-Tskhovrebova, 1960; Price & Brown, ...

Effects of Various Kynurenine Metabolites on Respiratory Parameters of Rat Brain, Liver and Heart Mitochondria

Previously, we demonstrated that the endogenous glutamate receptor antagonist kynurenic acid dose-dependently and significantly affected rat heart mitochondria. Now we have investigated the effects of L-tryptophan, L-kynurenine, 3-hydroxykynurenine and kynurenic, anthranilic, 3-hydroxyanthranilic, xanthurenic and quinolinic acids on respiratory parameters (ie, state 2, state 3), respiratory con...

Hankes and Henderson (I), Gholson et al. (2), and Gholson and Henderson (3) demonstrated that nL-tryptophan-7cu-14C was metabolized by the rat to r4C02 via acetate-1-r4C. Subsequently, Gholson et al. (4) indicated that 3-hydroxyanthranilic acid was an intermediate in the major pathway for the degradation of the benzene ring of tryptophan. Further experiments in viva have provided evidence that ...
